Utilization of DNA Fingerprinting for Identification of Grape (Vitis spp.) Cultivars

Abstract
The objective of this study was to assess genotype identification, parentage analysis, and the estimation of genetic relationship by DNA fingerprinting assay using genomic DNA of grape. DNA samples from 11 grape cultivars digested with Hinf I and Hae III and hybridized with simple repetitive (GGT)11 probe revealed resolvable complex polymorphic patterns. The probe detected the highest level of polymorphism of grape cultivars which were digested with only Hinf I endonuclease, with an average BS level of 55% between grape cultivars. Using the same probe, DNA samples digested with Hae III endonuclease had an average of seven specific bands and and an average of 52% BS. DFP patterns of grape cultivars revealed high polymorphism among cultivars and their progeny. Related cultivars such as ¢¥Black Olympia¢¥, ¢¥Pione¢¥, and ¢¥Benizuiho¢¥, which are probably progeny of ¢¥Kyoho¢¥, showed a relatively high level of BS (average BS among ¢¥Black Olympia¢¥, ¢¥Pione¢¥, ¢¥Benizuiho¢¥, and ¢¥Kyoho¢¥ was 80%). Genetic relationships among 11 grape cultivars were evaluated. Different probes gave different absolute d values. Rank correlations calculated between d values of different endonuclease enzymes were all higher than 0.7 (P $lt; 0.01). According to these results, it seems that ¢¥Black Olympia¢¥, ¢¥Pione¢¥ and ¢¥Benizuiho¢¥ were likely to be direct progeny of ¢¥Kyoho¢¥. ¢¥Campbell Early¢¥, ¢¥Super Hamburg¢¥, and ¢¥MBA¢¥ were progeny of ¢¥Muscat Hamburg¢¥. According to the d values evaluated in the current research, ¢¥Cheongsoo¢¥ was a progeny of ¢¥Himrod Seedless¢¥ and ¢¥Seibel 9110¢¥.
